Walter is perhaps best known these days for her portrayal of Lucille Bluth, the martini-loving, self-centered matriarch of the hapless Bluth family on the television show Arrrested Development, which ran from 2003 to 2006 and again in 2013 and 2018. Since 2009, Walter has also provided the voice of Malory Archer, mother of super-spy Sterling Archer (voiced by H. Jon Benjamin) and head of a dysfunctional intelligence agency on the adult animated sitcom Archer.

However, Walter's acting career goes back several decades. She started by acting on the Broadway stage, earning the Clarence Derwent Award for Most Promising Newcomer in 1963 for her role in the play Photo Finish. She starred in Clint Eastwood's 1971 directorial debut film, Play Misty For Me, and earned an Emmy award in 1975 for Amy Prentiss, in which she portrayed an investigator who becomes Chief of Detectives within the San Francisco Police Department, per Television Academy. Before her voiceover work on Archer, she provided the voice of Mama on the 1990s sitcom Dinosaurs.
